pDG Helper & Packaging plasmids for AAV production now exclusively available from PlasmidFactory

(PresseBox) (Bielefeld, ) PlasmidFactory has just received a worldwide exclusive license from the German Cancer Research Center (DKFZ) for manufacturing, distribution and use of the Helper & Packaging Plasmids from the pDG- respectively pDPfamily. "We have put the marketing of this system in the hands of our longterm cooperation partner. As an established supplier of research labs as well as biotech and pharmaceutical companies around the world, PlasmidFactory guarantees worldwide use of our advanced technology." explains Professor Dr. Jürgen Kleinschmidt from the department of tumor virology at DKFZ. The pDG system for the production of adenoassociated viruses (AAV) is a twoplasmid system. This means that in contrast to the tripletransfection with three different plasmids, only two components are needed: a plasmid with the gene to be transferred and the corresponding pDG plasmid that includes all the information for the growth and packaging of AAV vector plasmids (the socalled "helper & packaging" functions). An additional infection with helper viruses - as required with the original systems - is not necessary. "The pDG system is currently both in technical and economic terms the world's most efficient system for plasmid based AAV production. The plasmid production costs are reduced dramatically by the elimination of the third plasmid," says Dr. Martin Schleef, CEO of PlasmidFactory." The use of AAV vectors in gene therapy is currently increasing significantly and the pDGsystem makes the production process for these vectors much easier "adds Dr. Marco Schmeer, Product Manager R & D, PlasmidFactory. The pDG plasmids are manufactured in reproducible quality by using PlasmidFactory's standardized and controlled process. Through a comprehensive analysis a defined homogeneous product is guaranteed. All plasmids are fully characterized, including LAL test (LPSendotoxin), BCA test (total protein), UV spectroscopy, restriction analysis and the detection of RNA and chromosomal DNA with AGE. "The preparation of pDG plasmid DNA in qualities for GMP production of AAV is another integral part of our services," adds Dr. Schleef.
